finansowane przez Narodowe Centrum Nauki w ramach projektu OPUS 18 pt. „Identyfikacja i funkcjonalna charakterystyka mutacji mtDNA w nowotworach złośliwych gruczołu mlekowego u psów na podstawie badań genomicznych, epigenomicznych i proteomicznych”

Nazwa stanowiska: Stypendysta-Doktorant

Wymagania stawiane kandydatowi:

  1. Uczestnik studiów I, II stopnia lub uczestnik studiów doktoranckich, realizowanych w jednostkach naukowych i uczelniach wyższych na kierunku biotechnologia lub kierunku pokrewnym.
  2. Zainteresowanie pracą naukową w dziedzinach proteomika, biochemia białek, fizjologia zwierząt. Premiowany będzie udział w badaniach molekularnych, w szczególności proteomicznych dotyczących poszukiwania markerów stanów chorobowych u psów.
  3. Wiedza, umiejętności i doświadczenie w pracy badawczej z wykorzystaniem technik z zakresu biologii molekularnej i biotechnologii.
  4. Umiejętność obsługi aparatury badawczej wykorzystywanej w badaniach z zakresu genomiki i epigenomiki.
  5. Umiejętność obsługi programów bioinformatycznych w analizach z zakresu biologii molekularnej.
  6. Bardzo dobra znajomość języka angielskiego w mowie i piśmie umożliwiająca pisanie publikacji naukowych oraz prezentację referatów.
  7. Ogólne kompetencje – samodzielność naukowa, dyspozycyjność, bardzo dobra organizacja pracy, w tym umiejętność samodzielnej pracy, jak też pracy w interdyscyplinarnym zespole naukowym, kreatywne myślenie.

Udział w projekcie:

Stypendysta będzie uczestniczył w następujących zadaniach badawczych, których celem będzie identyfikacja biomarkerów prognostycznych i / lub diagnostycznych złośliwych guzów gruczołu mlekowego psów z wykorzystaniem technik proteomicznych, genomicznych i epigenomicznych, realizowanych w projekcie OPUS 18, pt. „IDENTYFIKACJA I FUNKCJONALNA CHARAKTERYSTYKA MUTACJI mtDNA W NOWOTWORACH ZŁOŚLIWYCH GRUCZOŁU MLEKOWEGO U PSÓW NA PODSTAWIE BADAŃ GENOMICZNYCH, EPIGENOMICZNYCH I PROTEOMICZNYCH”, realizowanym w ramach konsorcjum naukowego miedzy Uniwersytetem Przyrodniczym w Lublinie i Instytutem Genetyki i Biotechnologii Zwierząt Pan w Jastrzębcu,  realizowanym przez prof. dr hab. Brygidę Ślaską – kierownika projektu, oraz prof. dr hab. Mariusza Pierzchałę - kierownika zadań badawczych realizowanych w IGBZ PAN w Jastrzębcu.

  1. Identyfikacja biomarkerów guzów gruczołu mlekowego psów na podstawie różnic w proteomie w relacji do zmian w genomie mitochondrialnego DNA (mutacje, polimorfizmy i heteroplazmia) tkanki nowotworowej, prawidłowej i krwi.
  2. Ocena zmian profili proteomicznych oraz modyfikacji białek w oparciu o spektrometrię mas (Maldi TOF) w odniesieniu do zmian  mitochondrialnego DNA w tkance prawidłowej i nowotworowej gruczołu mlekowego psów.

Zakres obowiązków Stypendysty/Doktoranta:

  1. Prowadzenie badań naukowych przewidzianych w projekcie badawczym w zakresie proteomiki, genomiki i epigenomiki.
  2. Przygotowywanie prezentacji oraz manuskryptów dotyczących prowadzonych badań.
  3. Prezentacja wyników badań na konferencjach międzynarodowych.

Warunki zatrudnienia:

Aim of the project:

Major, unexplored difference between chromatin remodelling in fertilization and following nuclear transfer is the involvement of protamine in the former. Basing on that, the main aim of this project is to enhance cloning efficiency by replacing the histones in the somatic cell’s nucleus with protamine. The project aim is broken down in two Objectives. In objective 1 we will characterise in details somatic cell expressing protamine 1 (Prm1) as a new NR strategy, while in objective 2 we will verify the effect of new NR strategy on reprogramming efficiency.

Description of tasks:

Post-doc will be responsible for the protaminization of somatic cells, somatic cell nuclear transfer of sheep oocytes as well as molecular analysis (synthesis of the PEGP2-P2A-PRTM1vector; ChepSeq of protaminized cells; methylome and transcriptome analysis on single SCNT embryo and placenta).

    The Institute of Genetics and Animal Breeding of the Polish Academy of Sciences  (IGAB PAS)  in  Jastrzębiec n/Warsaw is seeking for scientists with expertise and hands-on experience inin molecular genetics / molecular biology / nutrigenomics and nutrigenetics.

    Candidates applying for the position should meet the following criteria:

    1. Degree of habilitated doctor in agricultural, biological, medical, veterinary or related sciences, and for candidates for full professors, the title of professor is required (with the exception indicated in Chapter 8, Art. 89. 1. Acts on the Polish Academy of Sciences of 30 April 2010 (Journal of Laws No. 96 item 619) );

    2. Have specific research interests supported by:

    • scientific achievements including original creative works, in particular publications in journals awarded by Journal Citation Reports;

    • high values ​​of scientific indicators: Hirsch index and the number of citations;

    3. Have documented experience in the above scope of research;

    4. Be familiar with adequate laboratory techniques;

    5. Direct or participate (in the past or currently) in a research project financed from external sources obtained through a competitive procedure;

    Project description:

    The HNF4A gene is one of an array of genes involved in organogenesis and in modulation of the cancer development process in liver. In particular, it is known in mice, which removal of this gene from the genome, leads to tumor development in liver. Hence, it is postulated that any loss-of-function variation to the gene structure/composition (mutation) can trigger liver cancer development and its progression. As far, a throughout functional analysis of the HNF4A mutations in human liver cancer have not been performed. This study will allow us to investigate for the first time the importance of the HNF4 gene mutations in liver cancer. We have already found that the HNF4 mutations are accumulated at the evolutionally sustained regions of the gene, which is partly the reason of our interest in those mutations. Since the HNF4A gene modulates liver tissue differentiation and liver cancer cell fate, the loss of its function is associated with a poor prognosis. Our study aims to demonstrate that pathogenic mutations of HNF4A cause dysregulation of several gene expressions and induce hepatic tumorigenesis. Therefore, the immediate gain in knowledge expected from this project is evaluation of the role of the HNF4A mutations in liver cancer onset. However, given the increasing information on single DNA units’ variants and alterations of their copy number stemming from the many cancer sequencing projects going on world-wide, there is strong need of functional validation to identify potential drivers – a pre-requisite to translate this knowledge into new diagnostic or therapeutic leads. For this reason, we perform functional study of HNF4A mutations as a model case. Our preliminary results show that the mutations of the HNF4A gene result in impaired gene functionality. We have also identified three mutations located within another region of the gene which all resulted in a reduction of the gene activity. Although we found that HNF4A mutations are possible pathogenic mutations, it is unclear how the HNF4A mutations impact the pathogenesis of liver cancer. To answer this question, we will introduce mutations using CRISPR-Cas9 system in mouse and human normal liver cell lines and subsequently perform functional assays. Gene mutations can lead to altered gene function and malignant cellular transformation. To improve the early detection of mutations, it is indispensable to have a complete map of cancer driver genes and their hot spots. Our study aims to support the ongoing efforts at improving the early detection of cancers, thereby contributing to the field of preventive medicine. Although the modern molecular tools have let us determine multiple mutations linked to several cancers, the data comes only from limited number of countries. Our study may help to identify pathogenic mutations without performing tedious analyses, but by using an already existing dataset.
    Description of the tasks:

    Task 1: To test whether Zn finger mutations of HNF4A cause dysregulation of transcriptional networks and induce hepatic tumorigenesis.

    Task 2: To investigate whether HNF4A ligand domain mutations cause liver cancer development through disrupting their binding specificity to target promoters and their co-factors, which changes the alterations in its transcriptional networks.
